The overall objective of P-CUBE is to provide more general access to European users to the most advanced techniques in cloning, expression, protein characterisation and crystallisation available in specialised laboratories. P-CUBE combines existing infrastructures and know-how of leading European laboratories in bacterial, eukaryotic expression of proteins, in high-throughput crystallization and in libraries design for the effective production and crystallization of macromolecules. To improve markedly current approaches to protein production and crystallisation, P-CUBE has the following main directions/aims:

  1. Provide an improved and automated technology for library based soluble construct screening
  2. Link soluble construct definition with crystallisation propensity,
  3. Partially automate the steps to select binding proteins for cocrystallisation,
  4. Automate eukaryotic protein expression with an emphasis on the parallel expression of
    proteins that form functional complexes
  5. Develop light microscopic techniques to overcome bottlenecks of automated eukaryotic expression.
