The overall objective of P-CUBE is to provide more general access to European users to the most advanced techniques in cloning, expression, protein characterisation and crystallisation available in specialised laboratories. P-CUBE combines existing infrastructures and know-how of leading European laboratories in bacterial, eukaryotic expression of proteins, in high-throughput crystallization and in libraries design for the effective production and crystallization of macromolecules. To improve markedly current approaches to protein production and crystallisation, P-CUBE has the following main directions/aims:
- Provide an improved and automated technology for library based soluble construct screening
- Link soluble construct definition with crystallisation propensity,
- Partially automate the steps to select binding proteins for cocrystallisation,
- Automate eukaryotic protein expression with an emphasis on the parallel expression of
proteins that form functional complexes
- Develop light microscopic techniques to overcome bottlenecks of automated eukaryotic expression.
